6月30日 ，强赛亞足聯官方公布世預賽亞洲區12強賽抽簽儀式安排，抽签从第抽簽從第六檔的档球队开档静敌球隊開始抽起 ，分別放置在A組和B組，始国以此類推直到抽完第一檔的足第球隊。
比賽為十個比賽日，候强分別為 ：2021年9月2日 、强赛9月7日，抽签从第10月7日 ，档球队开档静敌10月12日 ，始国11月11日 ，足第11月16日 、候强2022年1月27日 ，强赛2月1日，抽签从第3月24日、档球队开档静敌3月29日 。
兩個小組的前兩名直接晉級 ，兩個第三名將進行附加賽，勝者有資格參加最後洲際淘汰賽爭取世界杯參賽資格 。
根據6月18日國際足聯新一期的世界排名 ，12強賽參賽球隊分檔情況為 ：
第一檔 ：日本 、伊朗
第二檔 ：澳大利亞 、韓國
第三檔 ：沙特 、阿聯酋
第四檔：伊拉克 、中國
第五檔 ：阿曼 、敘利亞
第六檔：越南 、黎巴嫩
